Background ABI5 (abscisic acid insensitive 5) is involved in ABA-regulated gene expression during seed development and subsequent vegetative stage and acts as the major mediator of ABA repression of growth. Binds to the embryo specification element and the ABA-responsive element (ABRE) of the Dc3 gene promoter and to the ABRE of the Em1 and Em6 genes promoters.
ABI5 is a key component in ABA-triggered pathways during germination, seedling establishment, and vegetative growth (Lopez-Molina et al., 2001); in addition, it has been reported to play certain roles in nitrogen assimilation and signaling (Signora et al., 2001).

Required for ABI5 degradation, by mediating its ubiquitination. Together with EDR1, may regulate endocytic trafficking and/or the formation of signaling complexes on trans-Golgi network (TGN)/ early endosome (EE) vesicles during stress responses.

AT1G55660 : Comparison of seed and ABA-inducible vegetative gene expression in wild-type and abi5-1 plants indicates that ABI5 regulates a subset of late embryogenesis-abundant genes during both developmental stages. Responsible for reducing cadmium uptake, mediated by interaction with MYB49 . These data demonstrate that PKS5-mediated phosphorylation of ABI5 at Ser42 is critical for ABA regulation of seed germination and gene expression in Arabidopsis. It is expressed in seeds and up-regulated in response to ABA. Loss of function mutants show decreased rate of germination in the presence of ABA. ABA dependent regulation is mediated by both ABI3 and ABI5. ABI5 promotes MFT expression, primarily in the radicle-hypocotyl transition zone and ABI3 suppresses it in the seed.

Under these conditions, ABI4 and ABI5 each induced a small set of genes (95 and 59, respectively), with only 16 shared targets. In addition to effector genes involved in seed maturation and storage, several signaling proteins and transcription factors not previously known to be involved in the ABA signaling pathway were identified as targets of ABI4 and/or ABI5. negative regulation of seed germination Source: TAIR "The gibberellic acid signaling repressor RGL2 inhibits Arabidopsis seed germination by stimulating abscisic acid synthesis and ABI5 activity." ABI5-BINDING PROTEIN2 (AFP2) negatively regulates the abscisic acid signal by accelerating ABI5 degradation during seed germination in Arabidopsis ( Arabidopsis thaliana ). The abscisic acid signal is reported to delay flowering by up-regulating Flowering Locus C expression, but the role of AFP2 in regulating flowering time is unknown. Here, we found that flowering time was markedly delayed
ABI5 overexpression rescued ppc2 seedling growth arrest under low CO 2 conditions. It has been reported that ABI3, ABI4, and ABI5 (the downstream targets of the ABA signaling pathway) are required for ABA modulation of seed germination and post-germination development (Finkelstein and Lynch, 2000; Lopez-Molina et al., 2001). The abi5-5 mutation was characterized at the molecular level and was shown to result from a two base pair deletion in the coding sequence of the ABI 5 gene.
